Endogenous CSE/Hydrogen Sulfide System Regulates the Effects of Glucocorticoids and Insulin on Muscle Protein Synthesis

DEX inhibited protein synthesis, CSE expression and the mTOR/p70S6K signalling pathway. Myoblasts were incubated with different doses of DEX (0, 0.1, 1, or 10 μM) for 6 h. All cells were then incubated with puromycin (10 μM) for 30 min to measure the protein synthesis rates. Cell lysates were immunoblotted with specific antibodies. (a) Protein synthesis was measured with an antibody against puromycin, (b) CSE and CBS protein expression levels, (c) total mTOR and p-mTOR (Ser 2448) protein levels, and (d) p70S6K or p-p70S6K (Thr 389) protein levels.
