Endogenous CSE/Hydrogen Sulfide System Regulates the Effects of Glucocorticoids and Insulin on Muscle Protein Synthesis

H2S stimulated protein synthesis in chicken myoblasts. Myoblasts were incubated with or without NaHS (500 μM). Cells were simultaneously treated with or without DEX (10 μM) for 6 h. (a) The concentration of H2S in culture media supplemented with NaHS at different times (10, 30, 60 min, or 6 h). (b) Protein synthesis was measured with an antibody against puromycin. (c) CSE and CBS levels were detected by immunoblotting using CSE and CBS antibodies. (d) The total and phosphorylated mTOR levels were analysed with antibodies against mTOR and p-mTOR (Ser 2448), respectively; (e) the total and phosphorylated p70S6K levels were analysed using antibodies against p70S6K or p-p70S6K (Thr 389), respectively.
