Lupeol Counteracts the Proinflammatory Signalling Triggered in Macrophages by 7-Keto-Cholesterol: New Perspectives in the Therapy of Atherosclerosis

Western blotting analysis of autophagy markers in M(IFN-γ/LPS) macrophages. Lupeol enhances autophagy on M(IFN-γ/LPS) macrophages and counteracts dysregulated autophagy induced by 7-Keto-cholesterol (7KC). M(IFN-γ/LPS)-polarized THP-1 macrophages were stimulated or not with 25 μM lupeol for 1 hour and further stimulated with 7KC for 20 hours in the presence of the autophagy inhibitors ChQ or 3MA. Western blotting analysis of (a) LC3-I/II and (b) P62-SQSTM1 were performed in the whole-cell lysates of macrophages. Data are expressed relative to the control (fold increase) as of 4 independent experiments. Values mentioned are the ratio of LC-3-II to LC3-I. Symbols indicate significant differences tested by one-way ANOVA.
