Autophagy inhibits the activation of CHK1. (a, b) Autophagy activators reduce p-CHK1 accumulation induced by HU. Phosphorylation of CHK1 at S345 and total CHK1 were determined by Western blot analysis in wild-type and Rapa-treated or Torin1-treated cells at the indicated times following exposure to 200 μM HU. (c, d) Inhibition of autophagy leads to a significant upregulation of p-CHK1. (c) HT1080, U2OS cells, and MSCs were treated with CQ (50 μM) for 24 h for measurement of the protein level of p-CHK1 and total CHK1 by Western blot analysis.
