Glutamate Attenuates the Survival Property of IGFR through NR2B Containing N-Methyl-D-aspartate Receptors in Cortical Neurons

Glutamate inhibits IGF-1-induced activation of the PI3K/Akt pathway, while enhancing the effect of IGF-1 on ERK. Cultured cortical neurons were pretreated with glutamate (1 mM) for 1 h and then exposed to 100 ng/ml IGF-1 for 8 min. Determination of (a) AKT and PDK1 phosphorylation levels and (b) MEK and ERK1/2 phosphorylation levels by western blot. Blots represent prototypical examples of experiments replicated at least three times. .
