Application of MAPK and NF-κB pathway inhibitors. (a-d) Cells were preincubated with various concentrations of MAPK and NF-κB pathway inhibitors for 2 h and then treated with 50 μM PQ for 30 min. Cell viability assay was determined by CCK-8 assays. (e-h) Phosphorylation levels of p65, ERK, JNK, and p38 were detected by Western blot analysis. The PQ-induced phosphorylation of p65 was signally attenuated in the presence of the p65 inhibitor BAY (5 μM), while the phosphorylation of ERK, JNK, and p38 was observably reduced by the inhibitors SCH (2 μM), SP (10 μM), and SB (12.5 μM).
