Hyperoxia or NQO1 overexpression decreased 8-OHdG formation. Three stably transfected BEAS-2B cell lines Ctr, NQO1-NQO1, and SNP were incubated under RA or O2 for 48 h. Genomic DNA was isolated from the cells and digested with micrococcal endonuclease, spleen phosphodiesterase, nuclease P1, and calf intestinal phosphatase, followed with LC-MS of 8-OHdG using 0.2 μg enzyme-treated DNA (; ).
