The measurement of NOX and caspase activities. Cultured chondrocytes were pretreated with 10 mM NAC, 10 μM GW1929, 10 μM DPI, 10 μM SB203580, 10 μM PD98059, or 10 μM SP600125 for 1 h and then treated with 10 ng/ml IL-1β for 24 h. Chondrocytes were harvested, and total proteins were extracted, and the levels of NOX and cleaved caspases were detected by the assay kit: (a) NOX, (b) caspase-9, and (c) caspase-3. Results are presented as of three independent experiments.
