High-Fat Diet-Induced Fatty Liver Is Associated with Immunosuppressive Response during Sepsis in Mice

Palmitic acid-induced hepatocyte fat accumulation manifests as impaired immune competence to LPS challenge. (a) Murine primary hepatocytes were harvested from C57BL/6 mice and seeded in 12-well collagen I-coated plates and incubated in RPMI1640 medium. After attaching for 8 h, cells were washed three times with DPBS and stimulated with 0.4 mM PA or BSA control for 12 h. Then, the medium was replaced by the refresh medium to continue incubating for another 12 h. Finally, the supernatant was collected for further study. Raw264.7 cells were seeded at a confluence of 70-75% per well and treated with LPS (1 μg/ml) and hepatocyte supernatant for 24 h. (b) After incubating with PA or BSA, the TG content of primary hepatocytes was measured. BSA () and PA ().
