Acetylshikonin Induces Apoptosis in Human Colorectal Cancer HCT-15 and LoVo Cells via Nuclear Translocation of FOXO3 and ROS Level Elevation

Figure 6

Cell viability and antiproliferative effect of acetylshikonin-treated HCT-15 and LoVo cells were assessed when cells were cotreated with NAC in control and acetylshikonin (5 μM). (a) Cell viability was assessed with MTT assay. The data represent the of three independent experiments. Number sign indicates a significant difference of NAC-treated cells from acetylshikonin-treated cells (). (b) Control cells and acetylshikonin-treated cells were incubated for 24 h with or without NAC and stained with propidium iodide (PI) for a flow cytometric analysis of DNA content. Data are represented in bar graphs. (c) Apoptosis-related protein expressions were analyzed by western blot analysis. β-Actin was used as a gel-loading control.
