Cell viability assessed by MTT assay (a). The cells were pretreated with glucose (8 mM or 32 mM) for 15 min before undergoing a hypoxic insult for 24 h. For analysis of the effect of EHT 1864, cells were preincubated with EHT 1864 (20 μM) for 30 min, and after, they were incubated in the presence of glucose (8 and 32 mM). The data are presented as the mean ± standard deviation of three independent experiments. (b) Lysate proteins (1 mg/ml) were resolved by electrophoresis and analyzed by Western blot, using the antibodies against the indicated proteins. The band intensity of p-ERK and ERK was determined by densitometric analysis using the image software (c).
