Peroxisome Deficiency Dysregulates Fatty Acid Oxidization and Exacerbates Lipotoxicity in β Cells

Hypothesis schematic. A unifying hypothesis depicting the cascade of events in response to Pex14 KD that result in oxidative stress and programmed cell death in β cells. We speculate that Pex14 KD inhibits peroxisome biogenesis, which disturbs fatty acid oxidization, thus leading to lipid accumulation. Peroxisome deficiency also causes downregulation of catalase and Sod2, in turn H₂O₂ accumulation. H₂O₂ activates inflammatory factors and ERS, which in turn induces apoptosis and autophagy.
