Effect of Nrf2 knockdown on the protective effect of AR extract against Dox-induced toxicity in H9c2 cells. The H9c2 cells were transfected with scramble siRNA or siRNA against Nrf2 and pretreated with AR extract (1 mg/mL) or vehicle control (0.1% DMSO) for 12 h and then received a 0.5 μM Dox treatment for an additional 24 h. Cell viability and LDH levels in the transfected H9c2 cells were examined by (a) MTT and (b) LDH assays. (c) Intracellular ROS generation in H9c2 cells was detected by flow cytometry after CM-H2DCFDA staining. (d) Cell apoptosis in H9c2 cells was detected using flow cytometry after annexin V-FITC and PI double staining.
