MicroRNA-1224-5p Aggravates Sepsis-Related Acute Lung Injury in Mice

Figure 6

The miR-1224-5p antagomir prevents LPS-induced ALI through activating AMPKα. (a, b) Mice were intravenously injected with the agomir (30 mg/kg/day), antagomir (80 mg/kg/day), or matched controls for 3 consecutive days and then exposed to LPS (5 mg/kg) for 12 h. Phosphorylated and total AMPKα in lungs were detected. (c) To inhibit AMPKα, mice were intraperitoneally with CpC (20 mg/kg) once two days for 3 times before miR-1224-5p antagomir treatment, and then ROS and MDA levels in lungs were measured. (d, e) The levels of IL-1β, IL-6, IL-18, and TNF-α in murine lungs. (f) Lung wet to dry ratio. (g) LDH activities in lungs. (h) Total protein concentrations in BALF. (i, j) Pulmonary function was detected. (k) Arterial blood gas analysis of PaO2 and PaCO2. Data represent the ( per group). versus the matched group.
