Dopamine Cytotoxicity Involves Both Oxidative and Nonoxidative Pathways in SH-SY5Y Cells: Potential Role of Alpha-Synuclein Overexpression and Proteasomal Inhibition in the Etiopathogenesis of Parkinson's Disease

Accumulation of α-synuclein in SH-SY5Y cells after DA and lactacystin exposure. SH-SY5Y cells were incubated alone (control), with DA (400 μM), with lactacystin (5 μM), and with 400 μM DA co-treated with 2.5 mM NAC for 24 h. After 24 h of incubation, cell lysates were prepared and processed for Western blotting, as described in Section 3. The specificity of the commercially available antibody has been checked against pure recombinant human α-synuclein by Western blotting and immunodot blotting (data not shown).
