Cryptochrome and Period Proteins Are Regulated by the CLOCK/BMAL1 Gene: Crosstalk between the PPARs/RXR 𝛼 -Regulated and CLOCK/BMAL1-Regulated Systems

Dose-dependent alteration by the CLOCK/BMAL1 gene (0–220 ng) of the transcriptional activity of PPAR /RXR mediated through PPRE ( -AGGTCA- ) elements located in the promoter of acyl-CoA oxidase (AOX) in human kidney 293T cells in charcoal-treated medium. Alteration by the CLOCK/BMAL1 gene alone (a), plus the PPAR /RXR gene (50 ng) (b), and plus the PPAR ligand/activator fenofibric acid (10 M) (c) in activity of transcription by PPAR /RXR through PPRE ( -AGGTCA- ) elements located in the promoter of AOX. Luciferase activity for pAOX-Luc was normalized to the luciferase activity of pRL-TK. Experiments were performed in triplicate, and four independent experiments were performed. All data are means ± SD.
