A Dominant-Negative PPAR Mutant Promotes Cell Cycle Progression and Cell Growth in Vascular Smooth Muscle Cells

PPRE-mediated transcriptional activity of WT-, DN-, and CA-PPAR. NIH/3T3 fibroblasts were transiently transfected with a PPRE(3x) firefly luciferase reporter vector and a pRL-CMV renilla luciferase expression construct with or without plasmids expressing WT-, DN-, or CA-PPAR. After 24 hours of transfection, cells were treated with or without 10 M rosiglitazone (RSG). Luciferase activity was assayed 48 hours after transfection, adjusting for transfection efficiency by normalizing firefly luciferase activities to renilla luciferase activity. All transfections were performed a minimum of three times. Data are expressed as the ratios of firefly to renilla luciferase activity and presented as mean ± SE ( versus all other conditions by 1-way ANOVA).
