The Nitrated Fatty Acid 10-Nitro-oleate Diminishes Severity of LPS-Induced Acute Lung Injury in Mice
OA-NO2 activates PPAR- in airway epithelial cells. Binding of OA-NO2 to the PPAR- ligand-binding domain was modeled in silico. A549 cells were transiently transfected with pRLSV40 and with one of the following constructs: (c) PPAR-dependent luciferase reporter, pFATPluc; (d) PPAR- GAL4 reporter system (UASG 4 TK-luciferase + GAL4-PPAR-). Cells were then incubated with vehicle (DMSO), OA-NO2 (0.1–5 μM), or rosiglitazone (0.1–5 μM). After 24 h, luciferase activity was measured with a dual luciferase activity kit and normalized to that of Renilla luciferase.
