15-deoxy-Δ12,14-prostaglandin J2 Down-Regulates Activin-Induced Activin Receptor, Smad, and Cytokines Expression via Suppression of NF-κB and MAPK Signaling in HepG2 Cells

MEK and ERK protein expression in HepG2 cells. HepG2 cells were cultured with activin A (25 ng/mL) and 15d-PGJ2 (2 or 5 μM) for 72 h. The cells were lysed, and 30 µg of soluble protein was separated by performing electrophoresis on a 12% SDS-PAGE gel. Protein was detected by performing western blot analysis. The protein band intensities were quantified by using the i-image solution program, and the results are reported as the ratio of MEK (a) and ERK (b) to β-actin. The data represent the mean ± SD of 3 independent samples. * , comparison of stimulation with 15d-PGJ2 and activin A versus activin A stimulation alone.
