Chiglitazar Preferentially Regulates Gene Expression via Configuration-Restricted Binding and Phosphorylation Inhibition of PPARγ

Chiglitazar differentially affects promoter binding of PPARγ-containing transcription complexes. HPA-v cells were incubated with Chi (1 μM), Ros (1 μM), Pio (1 μM), and vehicle control (0.1% (vol/vol) DMSO) in the presence or absence of TNFα (5 ng/ml) for 24 hours. ChIP assays using anti-PPARγ antibody were then performed as described in Materials and Methods. The relative abundance of promoters bound to PPARγ was normalized against the input control sample.
