Fluid Retention Caused by Rosiglitazone Is Related to Increases in AQP2 and αENaC Membrane Expression

Figure 5

Effects of SR1664 and TNFα on PPARγ, p-PPARγ, AQP2, and αENaC in mIMCD-3 cells. (a, c, d) The effects of SR1664 on PPARγ, p-PPARγ, AQP2, and αENaC. (b) The changes of PPARγ and p-PPARγ upon coincubation with GW9662 (10 μM SR1664 + 5 μM GW9662). (e–g) The effects of TNFα on PPARγ, p-PPARγ, AQP2, and αENaC. The results show that SR1664 at 10 μM can significantly inhibit p-PPARγ; TNFα even 5 ng/ml can critically increase p-PPARγ, while there was no significant difference in the expression of AQP2 in cell membrane and cytoplasm and αENaC in membrane. After coincubation with GW9662 (10 μM SR1664 + 5 μM GW9662), the effect of SR1664 on p-PPARγ disappeared. The results are shown as mean ± s.e.m., . . . means compared to control; means compared to SR1664.
