Inhibitory Effects of a Novel PPAR-γ Agonist MEKT1 on Pomc Expression/ACTH Secretion in AtT20 Cells

Figure 3

Effects of MEKT1, rosiglitazone, and pioglitazone on ACTH secretion from AtT20 cells. (a) Effects of MEKT1, rosiglitazone, and pioglitazone on ACTH secretion into the media from AtT20 cells. AtT20 cells were treated with MEKT1, rosiglitazone (Rosi), and pioglitazone (Pio) (10 μM) or DMSO (0.1%) as a control. After 24-hour incubation of the cells, the ACTH secreted to the media was determined by EIA. (b) Dose-dependent effects of MEKT1 on ACTH secretion into the media from AtT20 cells. AtT20 cells were treated with MEKT1 (10 nM, 100 nM, 1 μM, or 10 μM) or 0.1% DMSO (vehicle control) for 24 hours. After 24-hour incubation of the cells, the ACTH secreted to the media was determined by EIA. Data are expressed as percentages (100%) of control. Each point represents mean ± SEM (). NS means “not significant.” , versus control.
