Stimulation of Neural Stem Cell Proliferation by Inhibition of Phosphodiesterase 5

Inhibition of the ERK1/2 pathway prevented proliferation of SVZ cells stimulated by PDE5 inhibitors. Cell proliferation following treatment with 1 μM U0126 and 1 μM T0156 (a, d), 1 μM sildenafil (b, e) or 10 μM zaprinast (c, f) was assessed by incorporation of EdU and evaluated by flow cytometry following 6 h and 24 h of treatment. Data are expressed as means ± SEM of at least 4 independent experiments. One-way ANOVA (Bonferroni’s post-test), *𝑃<0.05, **𝑃<0.01, and ***𝑃<0.001, significantly different from control and ⁺𝑃<0.05, and ⁺⁺⁺𝑃<0.001, significantly different from the PDE5 inhibitor.
