On the top, inversions rely on pLox sites that are used to invert the construct favoring other colors (Cre recombinase inverts segments floxed between two LoxP sites oriented in a head-to-head fashion). Both inverted and excised fragments can continue to invert as long as there is Cre activity, since LoxP sites are excised, while pLox sites are not. Consequently, an inducible model might be best suitable to stabilize cell color following inductor molecule removal. All recombinatorial events that do not involve the LoxP site upstream to the Stop cassette would result in no color acquisition by the cell.
