DCV does not impinge on cell proliferation or single cell clonogenicity. (a) A2780V cells were stained with 10 μM DCV (106 cells/mL) and SP/NSP fractions were flow sorted. Equal numbers of cells were then cultured and cell proliferation was assessed after 48 h using 3H-thymidine incorporation. Under these conditions, dye-retaining NSP cells proliferate equally well as their dye-extruded SP counterparts. (b) Comparative single cell cloning of freshly sorted (“+DCV”) versus presorted, in vitro expanded SP/NSP cells (“−DCV”). In two investigated cell lines (i.e., A2780 and IGROV1), no notable differences could be detected between the two experimental setups, indicating that the presence of DCV does not affect the clonogenic capacity of NSP cells.
