Wnt5a Increases Properties of Lung Cancer Stem Cells and Resistance to Cisplatin through Activation of Wnt5a/PKC Signaling Pathway

Figure 4

Wnt5a enhances the stemness of lung cancer cells determined by a clonogenic assay. A549 and A549/DDP cells were cultured with Wnt5a-CM with or without PKC inhibitor GF109203X; the capacity of clone formation was analyzed by using a clonogenic assay. (a) Representative images of clonogenic assay for A549 cells (left panel) and its relevant quantification of the total number of colonies from three 6-well plates (right panel). (b) Representative images of clonogenic assay for A549/DDP cells (left panel) and its relevant quantification of the total number of colonies from three 6-well plates (right panel). Wnt5a showed an ability to enhance clonogenic capacity in both A549 and A549/DDP cells. In contrast, GF109203X was able to inhibit the Wnt5a-enhanced clonogenicity in these cells. Compared with Control-CM group, represents . Data represented the mean ± SD from three independent triplicated experiments (). GF: GF109203X.
