MSCs and fd-ECM inhibit cellular gene expression in WHCO1 and MDA MD 231 cells. (a) Western blot analysis of lysates from WHCO1 cells cocultured with MSCs or cultured on an fd-ECM for 24 hrs showing expression of PCNA, cyclin D1, Bcl-2, p53, p21, MMP-2, MMP-9, and Bcl-xL. (b) Western blot analysis of lysates from MDA MB 231 cells cocultured with MSCs or cultured on an fd-ECM for 24 hrs showing expression of the same genes as in (a). Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was used as a loading control. ((c)-(d)) MMP-2 and MMP-9 gelatinolytic activities in WHCO1 (c) and MDA MB 231 (d) in media samples after 24 hrs of incubation at 37°C as described in Section 2. Gelatinolytic activity was observed as clear bands in the gels. Experiments were performed in triplicate and repeated at least 2 times.
