The Milk Thistle (Silybum marianum) Compound Silibinin Inhibits Cardiomyogenesis of Embryonic Stem Cells by Interfering with Angiotensin II Signaling

Figure 5

Effect of the procardiogenic agents FGF-2 (a) and EPA (b) on the inhibition of cardiomyogenesis by silibinin. Embryoid bodies were treated from day 3 to day 10 of differentiation with silibinin (20 μM) alone or with either FGF-2 (10 ng/ml) () or EPA (50 μM) () in the absence or presence of silibinin. On day 10, the number of contracting cardiac foci was counted. (c) Interference with PI3-K and NO signaling and its impact on the inhibition of cardiomyogenesis by silibinin. Embryoid bodies were treated from day 3 to day 10 with either silibinin (20 μM) alone or with either the PI3-K inhibitor LY294002 (5 μM) () or L-NAME (100 μM) () and the number of contracting cardiac foci was counted. , significantly different to the untreated control. , significantly different to the FGF-2 or LY294002-treated sample. n.s., not significant.
