miR-129-5p Regulates the Immunomodulatory Functions of Adipose-Derived Stem Cells via Targeting Stat1 Signaling

Figure 6

Enhanced expression of pStat1 in ASCs with miR-129-5p knockdown. (a) ASCs were isolated from C57BL/6 mice and were transduced with lentivirus containing GFP (NC) or miR-129-5p mimic. NC- and miR-129-5p mimic-transduced ASCs were treated with TNFα plus IFNγ for 12 h, and cells were collected in TRIzol. mRNA levels were determined with quantitative RT-PCR. NC- and miR-129-5p inhibitor-transduced ASCs were treated with TNFα plus IFNγ for 60 min, and pStat1 expression was examined by western blot assay (b) and immunofluorescence staining (c). Scale bar: 200 μm. NC- and miR-129-5p inhibitor-transduced ASCs were treated with TNFα plus IFNγ in the presence of fludarabine, mRNA levels were determined with quantitative RT-PCR (d), and iNOS protein levels were examined by western blot assay (e). and .
