lncRNA CYTOR Facilitates Osteogenic Differentiation of Human Periodontal Ligament Stem Cells by Modulating SOX11 via Sponging miR-6512-3p

Figure 5

CYTOR promotes osteogenic differentiation of PDLSCs through regulating SOX11. (a, b) Verification of the effect of knockdown on SOX11. The mRNA and protein levels of SOX11 was measured by qRT-PCR and western blot after silencing SOX11 by siSOX11. We used siSOX11-1 to silence SOX11 in the subsequent experiments. (c, d) qRT-PCR and western blot were conducted to detected the mRNA (c) and protein (d) levels of osteogenic related markers in PDLSCs after cotransfected with siSOX11 and overCYTOR or their negative controls followed by 14 days’ osteogenic differentiation. (e) ALP staining was performed. (g) ALP activity of indicated groups were detected after 7 days’ osteogenic induction. (f) Alizarin Red staining was performed to detect mineralized nodules formation of indicated groups. (h) Semiquantification of (f). , compared with the group as indicated.
