Stroke Status Evoked Adhesion Molecule Genetic Alterations in Astrocytes Isolated from Stroke-Prone Spontaneously Hypertensive Rats and the Apigenin Inhibition of Their Expression

The inhibitory effects of apigenin and pioglitazone on U937 cell adhesion in astrocytes isolated from SHRSP. The astrocytes isolated from SHRSP/IZM were cultured for 24 hours with or without 10, 30, or 50 μM of apigenin or pioglitazone and were then challenged with TNF-α (10 ng/mL). The U937 cells (1 × 10 6 cells/mL) were added and incubated with astrocytes at 37°C for 3 hours. The adhesion of U937 cells to the astrocytes was measured using quantitative monolayer adhesion assays, similar to those previously described [28]. The inhibitory effects of apigenin are indicated (fold). The number of bound U937 cells per square millimeter was determined by direct microscopy. The data are means ± S.D (n = 6).
