Lineweaver-Burk plot for the determination of the kinetic parameters of rainbow trout rhodanese. The enzyme was assayed at varying concentrations of Na2S2O3 at 50 mM KCN (a) and varying concentrations of KCN at 50 mM Na2S2O3 (b). The reaction mixture consisted of a 25 mM citrate-phosphate-borate buffer, pH 10.5, and 50 μL enzyme in a total volume of 1.75 mL at 25°C.
