Caspase-10 Is the Key Initiator Caspase Involved in Tributyltin-Mediated Apoptosis in Human Immune Cells

Caspase-10 inhibition prevents all TBT-induced apoptotic effects in Jurkat A3 cells. Jurkat A3 parental and the two deficient cell lines were analysed as shown in Figure 2 except the pretreatment was accomplished with zAEVD-fmk. Jurkat A3 wt cells were treated with 1 μM TBT or Fas-ligand (Fas-L) as positive control for 4 h, and the deficient cell lines were treated with TBT only before proteins were separated on SDS-gels and immunoblotted. A second set of samples was preincubated for 1 h with the caspase-10 inhibitor AEVD (8.7 μM) before TBT or Fas-L were added. Additionally, DNA fragmentation was recorded from the same samples after agarose gel electrophoresis.
