Time course and concentration dependency of uptake of AGE-2 and AGE-3 and their subcellular localization in macrophages. (a) The J774.1 macrophages were incubated with AGE-2 or AGE-3 (20 μg/ml) at 37°C for the indicated periods. The AGE-2 or AGE-3 in the cells was detected by specific polyclonal antibodies as in Figure 1. Representative images from three separate experiments are shown. The fluorescence intensities were quantified with the results shown representing means ± SEMs of three separate experiments. (b) The J774.1 macrophages were incubated with different concentrations of AGE-2 or AGE-3 at 37°C for 1 hr. Representative images from three separate experiments are shown. The fluorescent intensities were quantified and results are shown as means ± SEMs of three separate experiments.
